Quality Control: Oligonucleotide

Oligonucleotide syntheses are monitored by determining the trityl release following the first coupling and subsequent deprotection, and the last deprotection. The stepwise repetitive yield is computed from the length of the oligo and the fraction of the trityl groups remaining (final divided by initial). Oligos are rejected that do not have a repetitive yield >99%. The optical density at 260 nm of each purified oligo is measured and must be consistent with the trityl data. Random samples from each 96 well plate are subjected to capillary electrophoresis to monitor length heterogeneity.
